• the outer retina shows severe changes following horizontal cell ablation with diphtheria toxin (DT) treatment including loss of synaptic contacts and cone pedicle mosaic restructuring
• shape and arrangement of cone pedicles are changed following DT treatment; pedicles show atrophy and form complex clusters
• after DT treatment, many rod photoreceptors retract their terminals, while cone pedicles form clusters
• with horizontal cell ablation, many rod photoreceptors are lost, while cone number is not significantly affected
• at 8 weeks and 6 months after DT treatment, normal columnar alignment of photoreceptor nuclei is disrupted; about one third of photoreceptor nuclei are lost in treated mice
• after diphtheria toxin (DT) injection to retinal ablate horizontal cells in adult animals, the outer plexiform layer appears thinner at 2 weeks after treatment and is almost completely absent at 6 months
• under scotopic conditions, 6 months after treatment, a-wave amplitudes are significantly lower than in wild-type retina for intensities >0.5 cds/sq. meter and b-wave amplitudes are lower than wild-type for intensities >0.005 cds/sq. meter
• photopic electroretinograms show similar effects with the b-wave increasing with increasing light intensities in wild-type but not mutants
• ON responses (time to peak) are slower in retinae of treated mice while OFF responses are not significantly different, indicating that ganglion cell responses in treated mice are just mildly affected
• visual performance (visual acuity and contrast sensitivity) in DT-treated mutants is slightly lower than in wild-type despite morphology changes in outer retina
