132 animals from a (C57BL/6J x NOD/LtDOI)F2 intercross were typed for 162 microsatellite markers at an average spacing of 7.84 cM to identify QTLs associated with susceptibility to rheumatoid arthritis. (C57BL/6J x NOD/LtDOI)F2 animals were inoculated with serum from K/BXN T cell receptor transgenic mice to induce arthritis. Parental strain C57BL/6J is susceptible to serum transfer induced arthritis whereas NOD/LtDOI is resistant. Stia1 (serum transfer induced arthritis 1) maps to 81.6 cM on mouse Chromosome 1 with a LOD score of 4.5 at D1Mit33. The Stia1 interval spans 79 cM - 92. cM. Stia2 maps to 23.5 cM on mouse Chromosome 2 with a LOD score of 7 at the Hc (hemolytic complement) locus. The Stia2 locus spans 11 cM - 41.4 cM. Hc is a possible candidategene for Stia2 because Hc appears to be required to develop arthritis via serum transfer, and Hc is known to be defective in the NOD/LtDOI strain.
