| Caption | (A) Summary of defects observed in E9.5 and E10.5 Bltp2-/- placentas. The patterned overlay indicates the phenotype is observed in at least half but not all KO placentas examined. (B) Histological images of adjacent sections from a representative E9.5 Bltp2-/- placenta demonstrating normal development comparable to controls (n = 3). Dashed boxes indicate the LZ region shown at higher magnification in C. While the MCT1 and MCT4 layers appear slightly delayed compared to controls in the KO placentas shown, this phenotype is not consistent across all KO placentas examined. (D) Images of the embryonic component of the conceptuses shown in B and C. (E) Histological images of adjacent sections from a representative E10.5 Bltp2-/- placenta displaying a diminished Tpbp+ SpT layer and moderate LZ defects relative to controls. Dashed boxes indicate the LZ region shown at higher magnification in F. While the SpT layer abnormality is observed in all four KO placentas examined, LZ defects are not fully penetrant. However, the severity of LZ defects correlates with the severity of the embryonic phenotype. In one KO placenta, the LZ develops normally. In contrast, the Tfeb+ and MCT1+ SynTI region is markedly reduced in 2 out of 4 KOs, and Gcm1/MCT4+ SynTII development/ chorioallantoic branching is impaired in 3 KOs. Fetal and maternal blood are present in the LZ of most KO placentas; however, discrete blood spaces are less abundant than in controls (n = 3/4). (G) Images of the embryonic component of the conceptuses shown in E and F. (H) Examination of additional KO placentas demonstrates that a thinner Pl-1+ TGC layer accompanies the diminished Tpbp + SpT layer (n = 3). Scale bars: 500 m. |
