| Caption | (A) Summary of defects observed in E9.5 and E10.5 Arpc1a-/- placentas. The patterned overlay indicates the phenotype is observed in at least half but not all KO placentas examined. (B) Histological images of adjacent sections from a representative E9.5 Arpc1a-/- placenta demonstrating normal development comparable to controls (n = 3). Dashed boxes indicate the LZ region shown at higher magnification in C. (D) Images of the embryonic component of the conceptuses shown in B and C. (E) Histological images of adjacent sections from a representative E10.5 Arpc1a-/- placenta displaying an expanded Tpbp+ SpT layer and multiple LZ defects relative to controls (n = 3). Dashed boxes indicate the LZ region shown at higher magnification in F. In KO placentas, the Tfeb+ and MCT1+ SynTI region is markedly reduced compared to controls. The SynTII layer/ chorioallantoic branching is nearly absent, as Gcm1 and MCT4 expression is sparse and remains at the chorioallantoic interface. Although some endothelial cells and fetal blood cells are detected among labyrinth trophoblasts, discrete fetal blood spaces are noticeably decreased. Large pools of maternal blood are observed adjacent to the LZ; however, integrated discrete maternal blood spaces are also reduced. Although MCT1 and the more severely diminished MCT4 signals are found in close apposition, the extensive network of adjacent MCT1+ SynTI and MCT4+ SynTII cells observed in controls is absent. (G) Images of the embryonic component of the conceptuses shown in E and F. (H) Examination of additional KO placentas (n = 3) reveals a normal Pl-1+ TGC layer and normal Tpbp+ SpT layer, in contrast to the previously examined KO placentas exhibiting an expanded SpT layer. Scale bars: 500 m. |
