| Caption | (A) Summary of defects observed in E9.5 Got2-/- placentas. (B) Histological images of adjacent sections from a representative E9.5 Got2-/- placenta displaying a diminished Tpbp+ SpT layer, abnormal chorioallantoic interface, and multiple LZ defects relative to controls (n = 3). Dashed boxes indicate the LZ region shown at higher magnification in C. An abnormal chorioallantoic interface is observed in KO placentas, characterized by a reduction of allantoic mesenchyme. The Tfeb+ and MCT1+ SynTI region fails to expand and remains a thin layer compared to controls. The absence of invaded allantoic mesenchyme and minimal Gcm1/MCT4 expression indicate a complete failure of chorioallantoic branching. Endothelial cells and fetal blood cells remain confined to the allantoic mesenchyme. Maternal blood similarly fails to integrate among labyrinth trophoblasts and instead accumulates between the JZ and LZ. Although the minimal MCT1 and MCT4 signals are found in close apposition, the extensive network of adjacent MCT1+ SynTI and MCT4+ SynTII cells observed in controls has not developed. (D) Images of the embryonic component of the conceptuses shown in B and C. (E) Examination of additional KO placentas demonstrates that a thinner Pl-1+ TGC layer accompanies the diminished Tpbp+ SpT layer (n = 3). Scale bars: 500 m. | |||||||||||||||||||||||||||||||||||||||||||||||||||||||||||||||
