Mice homozygous for a null allele show accelerated liver tumor formation in DEN/CCl4-induced and diet-induced hepatocarcinogenesis. Mice homozygous for a different null allele exhibit impaired glucose tolerance, insulin resistance, accumulation of hepatic fat and acetyl-CoA, and increased de novo lipogenesis and cholesterol biosynthesis in the liver, independent of a high-fat diet.
