    A new BSB line containing the Lipctm1Unc mutation was generated by backcrossing (B6.129P2-Lipctm1Unc/J x SPRET/Ei)F1 animals back to the B6.129P2-Lipctm1Unc/J parent. Authors refer to this congenic as BSBHLKO (BSB hepatic lipase knockout.)

    A previous study detected linkage to obesity on mouse Chromosome 2 so markers on this chromosome were screened in BSBHLKO mice to identify QTLs. Significant linkage to fat pad weight, body mass index (BMI), and adiposity index (AI) was observed between markersD2Mit106 (75.6 cM) and D2Mit109 (81.7 cM) with peak LOD=3.7. This locus is named Bsbob (BSB obesity). SPRET/Ei-derived alleles confer decreased fat pad weight and mass and decreased BMI and AI with an additive or dominant mode of inheritance. The Lipc mutation is not linked to the phenotype.

    The effect of Bsbob was confirmed in a congenic carrying SPRET/Ei-derived DNA between D2Mit194 (81.4 cM) and D2Mit311 (83.1 cM) on a C57BL/6J genetic background. This congenic, B6.S-D2Mit194-D2Mit311, exhibits a lean phenotype. The estimated size of the congenic region is a minimum of 26.7 Mb to a maximum of 32.1 Mb.

    Bsbob colocalizes with three previously mapped obesity-related QTLs, Mob5, Mob6, and Mob7. Candidate genes mapping near Bsbob are a (agouti), Lpin3, Pltp, Acss1, Pcsk2, Sstr4, E2f1, Ghrh, and 2610042O14Rik. The region of Bsbob is syntenic to human Chromosome 20.
