147 SSLP markers were screened in a (C57BL/6J x C3H/HeJ)F2 intercross population to identify QTLS associated with particle-induced immune dysfunction. Animals were exposed to acid-coated carbon particles (ACP) and assessed for alveolar macrophage function. Parental strain C57BL/6J is susceptible to particle-induced immune dysfunction and exhibits decreased macrophage phagocytic function 1, 3, and 7 days after ACP exposure. Parental strain C3H/HeJ is fairly resistant. A locus showing significant linkage to this trait, Pidys, mapped to an interval on mouse Chromosome 17 between D17Mit147 (18.1 cM) and D17Mit64 (22.8 cM). Candidate genes in the region of Pidys are Hspa1b, Hspa1a, Tnf, Mcpt6, Mcpt7, and several histocompatibility genes. Pidys co-localizes with a previously identified QTL for ozone-induced bronchial hyperresponsiveness, Bhr3. A suggestive locus associated with particle-induced immune dysfunction was also identified on mouse Chromosome 11 mapping to approximately 47 cM.
