| Caption | (A) Summary of defects observed in E9.5 and E10.5 Pgm1-/- placentas. (B) Histological images of adjacent sections from a representative E9.5 Pgm1-/- placenta displaying multiple LZ defects relative to controls (n = 3). Dashed boxes indicate the LZ region shown at higher magnification in C. In KO placentas, the Tfeb+ and MCT1+ SynTI region is markedly reduced. The SynTII layer/chorioallantoic branching is also impaired compared to controls, demonstrated by the reduced Gcm1/MCT4+ network. Although MCT1 and the more severely diminished MCT4 signals are found in close apposition, MCT1+ SynTI and MCT4+ SynTII cells are rarely in close apposition and do not form the extensive adjacent network observed in controls. Although some endothelial cells and fetal and maternal blood cells are detected among labyrinth trophoblasts, discrete blood spaces are noticeably decreased. (D) Images of the embryonic component of the conceptuses shown in B and C. (E) Histological images of adjacent sections from a representative E10.5 Pgm1-/- placenta displaying multiple LZ defects relative to controls (n = 4). Dashed boxes indicate the LZ region shown at higher magnification in F. At E10.5, the Tfeb+ and MCT1+ SynTI region is still markedly reduced. SynTII layer development/ chorioallantoic branching is nearly absent, as Gcm1 and MCT4 expression is sparse. Although some endothelial cells and fetal blood cells are detected among labyrinth trophoblasts, discrete fetal blood spaces are noticeably decreased. Although MCT1 and the more severely diminished MCT4 signals are found in close apposition, the extensive network of adjacent MCT1+ SynTI and MCT4+ SynTII cells observed in controls is absent. (G) Images of the embryonic component of the conceptuses shown in E and F. Scale bars: 500 m. |
