| Caption | (A) Summary of defects observed in E9.5 and E10.5 Stt3b-/- placentas. (B) Histological images of adjacent sections from a representative E9.5 Stt3b-/- placenta displaying multiple LZ defects relative to controls (n = 3). Dashed boxes indicate the LZ region shown at higher magnification in C. In KO placentas, while the Tfeb+ region is relatively comparable to controls, the MCT1+ SynTI region is drastically reduced. However, Tfeb signal intensity appears elevated, suggesting a higher density of Tfeb+ cells. While this phenotype appears mild in the representative placenta shown, it is more pronounced when comparing all three KOs directly to the three controls. The SynTII layer/ chorioallantoic branching is also severely impaired, demonstrated by the reduced Gcm1/MCT4+ network. While some endothelial cells and fetal and maternal blood cells are detected among labyrinth trophoblasts, discrete blood spaces are noticeably decreased. Although MCT1 and MCT4 signals are found in close apposition, the extensive network of adjacent MCT1+ SynTI and MCT4+ SynTII cells observed in controls has not developed. (D) Images of the embryonic component of the conceptuses shown in B and C. (E) Histological images of adjacent sections from a representative E10.5 Stt3b-/- placenta displaying multiple LZ defects relative to controls (n = 3). Dashed boxes indicate the LZ region shown at higher magnification in F. In KO placentas, the Tfeb+ labyrinth trophoblast layer is slightly reduced compared to controls. However, Tfeb signal intensity within this domain appears elevated, suggesting an increased density of Tfeb+ cells despite the overall reduction in layer thickness. While this phenotype appears mild in the representative placenta shown, it is more pronounced when comparing all three KOs directly to the three controls. The SynTII layer/ chorioallantoic branching is also severely impaired, demonstrated by the reduced Gcm1/MCT4+ network. While endothelial cells and fetal and maternal blood cells are detected among labyrinth trophoblasts, discrete blood spaces are noticeably decreased. Although MCT1 and MCT4 signals are found in close apposition, the extensive network of adjacent MCT1 SynTI and MCT4 SynTII cells observed in controls is absent. (G) Images of the embryonic component of the conceptuses shown in E and F. Scale bars: 500 m. |
