| Caption | (A) Summary of defects observed in E9.5 Slc30a9-/- placentas. The patterned overlay indicates the phenotype is observed in at least half but not all KO placentas examined. (B) Histological images from representative E9.5 Slc30a9-/- placentas, displaying impaired chorioallantoic attachment and multiple LZ defects relative to controls (n 3). Due to the drastically small size of KO embryos and concern about having enough sections around the midline to distribute for separate histological analysis, all sections from a KO or control placenta were subjected to a single assay. Dashed boxes indicate the LZ region shown at higher magnification in C and D. Failure of chorioallantoic attachment is observed in most KO conceptuses examined (n = 8/14). However, in the conceptuses with normal attachment (n = 6/14), less allantoic mesenchyme is observed attached to the chorion compared to controls. In the KO placentas, the Tfeb+ region fails to expand and remains a thin layer compared to controls. While the MCT1+ SynTI cell layer has expanded, it remains thinner compared to controls. Gcm1 and MCT4 are restricted to the basal chorion and KO placentas with proper chorioallantoic attachment exhibit only shallow invasion of allantoic mesenchyme. This demonstrates that while chorioallantoic branching and SynTII differentiation initiate in these KOs, further elongation and secondary branching of these Gcm1/MCT4 SynTII outlined projections fail. Endothelial cells and fetal blood cells remain confined to the shallow projections of allantoic mesenchyme. Maternal blood similarly fails to integrate among labyrinth trophoblasts and instead accumulates between the JZ and LZ. Although MCT1 and the more limited MCT4 signals are found in close apposition, the extensive network of adjacent MCT1+ SynTI and MCT4+ SynTII cells observed in controls has not developed.
