| Caption | (A) Summary of defects observed in E9.5 Gcn1-/- placentas. The patterned overlay indicates the phenotype is observed in at least half but not all KO placentas examined. (B) Histological images of adjacent sections from a representative E9.5 Gcn1-/- placenta displaying impaired chorioallantoic attachment and multiple LZ defects relative to controls (n = 4). The Tfeb probe overstained during color development, resulting in lighter artifact staining adjacent to the true, intensely dark signal. Dashed boxes indicate the LZ region shown at higher magnification in C. Impaired chorioallantoic attachment is observed in most KO conceptuses examined (KO #2, n = 3/4). Although allantoic mesenchyme is present adjacent to the chorion, the attachment is weak and easily disrupted during dissection. In the conceptus with normal attachment (KO #1, n = 1/4), less allantoic mesenchyme is observed, and the resulting interface fails to expand as widely as in controls. In the KO placentas, the Tfeb+ and MCT1+ SynTI region fails to expand and remains a thin layer compared to controls. Gcm1 and MCT4 are restricted to the basal chorion and only shallow invasion of allantoic mesenchyme is observed. This demonstrates that while chorioallantoic branching and SynTII differentiation initiate, further elongation and secondary branching of these Gcm1/MCT4 SynTII outlined projections fail. Endothelial cells and fetal blood cells remain confined to the shallow projections of allantoic mesenchyme while maternal blood similarly fails to integrate among labyrinth trophoblasts and instead accumulates between the JZ and LZ. Likewise, the small number of MCT1+ SynTI and MCT4+ SynTII cells are not found in close apposition, in contrast to the extensive network of adjacent SynTI/SynTII cells observed in controls.
