Mutation description

• Mutation details: The transposon containing an unspecified hybrid actin promoter driving expression of red fluorescent protein (RFP) flanked by piggyBac-specific left and right terminal sequences was co-injected with a piggyBac-containing transgene to mobilize the transposon. The transposon DNA fragment containing RFP sequence was inserted in the 15th intron of the gene. At E9.5, qPCR analysis showed that mRNA levels were reduced by nearly 100% and by ~50% in homozygous and heterozygous embryos, respectively. Western blot analysis demonstrated that the transposon insertion resulted in significant truncation of the protein. Native protein expression was reduced to ~50% in heterozygous embryos and absent in homozygous embryos. qPCR and immunofluorescence confirmed that mRNA expression in heterozygous kidneys was ~50% of that in wild-type kidneys.
