Mutation details: This transgene contains the "CAG promoter" -- consisting of the CMV-IE enhancer followed by the chicken beta-actin (ACTB) upstream promoter, first exon and first intron -- driving expression of a loxP-flanked lacZ/neoR fusion gene (betageo) with three copies of the SV40 polyadenylation signal (3xSV40p[A]). Upon excision of betageo by Cre recombinase, the tetracycline transactivator (tTA) and enhanced green fluorescent protein (EGFP) genes are expressed from the CAG promoter. Presence of the tTA protein promotes expression of other transgenes regulated by the tet operon (tetO); this expression is readily suppressed by doxycycline. (J:153278)
