Mutation details: Site-directed mutagenesis converted the TGT codon to TTT resulting in the amino acid substitution of phenylalanine for cysteine at position 305 (C305F). Additional codons were modified to introduce an SpeI site. A floxed neo cassette was removed by cre mediated recombination. The amino acid substitution disrupts binding to Smr3a (RPL5) and RPL11.
