Mutation details: A threonine to arginine substitution was introduced into amino acid 61 (T61R) of mouse Apoe exon 3 and a floxed neo cassette was inserted into the downstream intron via homologous recombination. This mutation allows the mouse protein to mimic human APOE4 domain interaction. The presence of the neo cassette renders this allele hypomorphic with mRNA expression at 2%-5% that of wild-type. Expression of the mutant allele was observed by Western blot analysis. (J:75681)
