Mice homozygous for a knock-out allele show pre-implantation lethality associated with failure of embryo implantation and ectopic expression of an epiblast marker in the trophectoderm, indicating defects in trophoblast specification. Cultured blastocysts fail to hatch from the zona pellucida with apparent cell death and form atypical outgrowth colonies with no trophectoderm, primitive endoderm or epiblast cells.
