Mutation details: This transgene contains the mouse 8.6-kb calcium/calmodulin-dependent kinase II alpha promoter, the longest MAPT isoform, human tau441, cDNA with partial sequences of MAPT introns 9 and 10 inserted into the corresponding sides of exon 10 with a C to T mutation at position +16 (intron 10 + 16C to T) relative to the 5' splice site (predicted to lie within the stem) in intron 10 which increases splicing of exon 10. This line expresses tau protein levels of 186% of those in non-transgenic littermates, representing levels of human tau of approximately 86% of the levels of endogenous mouse tau. RT-PCR shows that mice exclusively express mRNA without exon 10 at 7 days of age and mRNA with and without exon 10 at a ratio of approximately 1:1 at 4 months of age. Quantification of signals of the alkaline phosphatase-treated samples show that mice predominately expresses 4R human tau (4R/3R ratio of 12.1) indicating altered splicing of exon 10 to increase 4R tau expression at an adult age.
