Quantitative Trait linkage analysis by LTC-IC (Long-term culture-initiating cell analysis) of 26 BXD RI (C57BL/6J x DBA/2) lines suggested that two loci have major quantitative effects on hematopoietic stem cell frequencies and are located on Chromosome 1 near Adprp (Scfr1) and Acrg (Scfr2) respectively.

This study focuses on one of these loci,Scfr1. B6.C-H35 mice (having a C57BL/6 background and parts of Chromosome 1 from BALB/c) demonstrated stem cell numbers intermediate to those of C57BL/6 (having lower numbers) and BALB/c (having higher levels) implying that an involved gene mapped to a Chromosome 1 region. SSLP's (Simple Sequence Length Polymorphism's) were used to map this region, in the B6.C-H35 and B6.C-H25 mice. B6.C-H35 mice had stem cell levels similar to the C57BL/6 mice and carried the C57BL/6 alleles at all locitested (D1Mit215, D1Mit365, D1Mit26 , D1Mit218, D1Mit33, D1Mit452, D1Mit113, D1Mit166, D1Mit17, and D1Mit155) while the B6.C-H25 typed like BALB/c are D1Mit113, D1Mit166, and D1Mit17 and like C57BL/6 at the other loci. The BALB/c donated region extends at least from D1Mit113 to D1Mit17, containing Apoa2, Ly9, Apcs, H25 and Scfr1. Thus, Scfr1 was suggested to map in the neighborhood of Adprp on mouse Chromosome 1.
