Linkage analysis was performed on 269 animals from a (HIII x LIII)F2 intercross to identify QTLs associated with pristine-induced arthritis phenotypes. Pristane injections were given intraperitoneally and animals were observed weekly for 200 days. Parental strain LIII is susceptible to pristine-induced arthritis, showing signs of disease beginning at 45 days post-inoculation and reaching 100% incidence after day 105.

Linkage to pristine-induced arthritis onset (LRS=14.7; 5% of variance), severity (LRS=19.9; 7% of variance), and incidence (LRS=13.7; 5% of variance) mapped to 45.8 cM on mouse Chromosome 3 near D3Mit100. This locus is designated Prtia1 (pristane induced arthritis 1) and appears to have an additive effect. Prtia1 colocalizes with previously identified arthritis QTLs Pgia26 (52.5 cM) and Cia5 (40 cM). Potential candidate genes mapping to this region include Fcgr1 (45.2 cM), Il6ra (42.1 cM), Cd53 (50.5 cM), Ctsk (47.9 cM), and Ctss (42.7 cM).

Suggestive linkage to pristine-induced arthritis incidence mapped to 29 cM on mouse Chromosome 6 near D6Mit123 (LRS=5.4; 2% of variance), and suggestive linkage to severity mapped to 58 cM on mouse Chromosome 12 near D12Mit19 (LRS=5.8; 2% of variance.)

Suggestive linkage to pristine-induced arthritisseverity in males mapped to 9 cM on mouse Chromosome 9 near D9Mit90 (LRS=6.5; 4% of variance), and suggestive linkage to incidence in males mapped to 92.3 cM on mouse Chromosome 1 near D1Mit35 (LRS=5.7; 4% of variance.)
