  • Experiment
    Authors used an intersubspecific backcross to localize sf to a 0.3 cM interval on Chromosome X. A BAC contig was constructed across the interval and 4 BACs 196K10, 34916, 8C22 and 284K10 defined a minimal tiling path over the estimated 500kb region. Computational analysis of the sequence from this region corresponded well with a transcript map generated in J: 62168. The order and orientation of markers in this region was determined to be: centromere - DXCch1 - DXMit55 - AA273854 - 3' - Ppp1r3f - 5' - 5'-Foxp3 - 3' - 3' - DXImx40e - 5' - 5' - Cacna1f - 3' - DXMit26 - 5' - Syp - 3' - 5' - Lmo6 - 3' - 3' - Plp2 - 5' - 3' - Pdzx - 5' - 5' - DXImx41e - 3' - 5' - Ppia-ps - 3' - 5' - Wdr45 - 3' - 5' - DXImx39e - 3' - 3' - AA030924 - 5' - 5' - Smt3h2-ps - 3'- (5' - Tcfe3 - 3' - DXCch4) - 5' - Gripap1 - 3' - 5' - Kcnd1 - 3' - 5' - Otud5 - 3' - DXCch2 - 5' - Pim2 - 3'. Candidates for the sf mutation were analyzed by direct sequencing of RT-PCR products from mutant and normal animals. A frameshift resulting in atruncated gene product was observed in the Foxp3 gene of sf animals. The identity of the Foxp3 gene was confirmed by the use of transgenic animals.

    DXCch5 also mapped to mouse Chr. X was was not shown in the contig.
