    Using differential display analysis, the authors were able to identify a cDNA fragment expressed in control but not mutant testis. This fragment hybridized to a ~15kb mRNA. Southern blot-RFLP analysis with the Hst6.7b fragment demonstrated its mapping within <0.2 cM of the Ccua locus. The Dnahc8 gene was also previously mapped in the vicinity of the Ccua locus. High resolution mapping using the Hst6.7b and Dnahc8 P1-loop probes on mice carrying various combinations of S-+ recombinant Chromosome 17 homologs revealed the co-localization of both genes at the Ccua locus. The Dnahc8 P1-loop probe also hybridized to ~15kb fragment. This suggested that both probes were different ESTs from orthologs of the same gene. A physical map of the region from 16.775 cM to 17.175 cM on Chromosome 17 was constructed using a contig of nine BACs (99D17, 324M10, 98H21, 351H18, 138O17, 283H8, 173G10, 566K2, 300K16).

    Southern blot analysis of restriction digested BAC clones and BAC-end sequence analysis demonstrated that the distance between Hst6.7b and Dnahc8-P1-loop was no more than 65kb in the genome and Hst6.7b might represent the 3' end of the message. The hybridization pattern also indicated that Hst6.7b and Dnahc8-P1 loop might represent sequences from the same gene. The order of markers on mouse Chromosome 17 is inferred to be: Pim1 - D17Mit61 - 3' Dnahc8 - 5' - D17Mit81 - D17Mit135 - Tff3 - D17Mit146 - D17Mit191 - Cryaa.
