Caption Generation of the Crisp4tm1.1Mkob (Flox allele without Neo) and Crisp4tm1.2Mkob (KO) alleles. The Crisp4 flox allele was generated by the insertion of a loxP-NeoR-loxP cassette in intron 3 and a third loxP site in intron 2 of the mouse Crisp4 gene, through homologous recombination. The targeting cassette was flanked by a ~5-kb 5' homology arm covering the sequence of intron 1-exon 2-intron 2 and a ~5-kb 3' homology arm covering the sequence of intron 3-exon 4-intron 4. The NeoR gene was subsequently removed by low dose of Cre recombinase activity (partial Cre) which generates the Crisp4tm1.1Mkob allele. Full Crisp4 knockout mice were generated by crossing heterozygous flox allele transgenic mice carrying Cre recombinase under the CMV promoter, generating the Crisp4tm1.2Mkob allele. The location of the F1 and R1 primers used for genotyping are indicated by the arrows.
