Caption Generation of the Nkx2-2tm4.1(cre/EGFP)Suss allele. Nkx2-2LCA is the RMCE acceptor allele integrated into 4H9 ES cells (Arnes et al., 2012). RMCE lox sites are indicated by colored arrows. The targeting vector includes an Frt-flanked hygro selection cassette. The cre/EGFP gene was recombineered in-frame at the Nkx2-2 ATG codon and deleted most of coding exon 1. Ex I' signifies the truncated region of coding exon 1 and Ex II refers to the coding exon 2. The cre/EGFP cassette replaces the LCA allele using RMCE. P1 and P2 = 5' PCR primers that verify proper integration of the hygromycin cassette. P3 and P4 = 3' primers that distinguish the wild-type allele and the newly integrated cre/EGFP allele with the 2722 lox site. Flpe recombinase removed the Frt-flanked selection cassette. P5 and P6 = primer set used to identify the cre/EGFP allele.
