Caption Placental phenotype in Stt3bem1(IMPC)J/Stt3bem1(IMPC)J (Stt3b-/-) embryos. At E9.5, Stt3b-/- placentas display multiple labyrinth zone (LZ) defects compared to controls. In KO placentas, chorioallantoic branching is impaired compared to controls. Although the Tfeb+ labyrinth trophoblast layer is comparable to controls, signal intensity appears elevated, suggesting a higher density of Tfeb+ cells. While this phenotype appears mild in the representative placenta shown, it is more pronounced when comparing all three KOs directly to the three controls. At E10.5, Stt3b-/- placentas exhibit multiple LZ defects compared to controls. In KO placentas, the Tfeb+ labyrinth trophoblast layer is slightly reduced in thickness but the Tfeb signal intensity appears elevated, suggesting an increased density of Tfeb+ cells. While this phenotype appears mild in the representative placenta shown, it is more pronounced when comparing all three KOs directly to the three controls. Chorioallantoic branching is also severely impaired.
