| Caption | At E9.5, Gcn1em1(IMPC)J/Gcn1em1(IMPC)J (Gcn1-/-) placentas exhibit impaired chorioallantoic attachment and multiple labyrinth zone defects compared to controls. The Tfeb probe, overstained during color development, resulting in lighter artifact staining adjacent to the true, intensely dark signal. In the KO placentas, the Tfeb+ region fails to expand and Gcm1+ chorioallantoic branching is severely impaired. Impaired chorioallantoic attachment is observed in most KO conceptuses examined (KO #2, n = 3/4). Although allantoic mesoderm is present adjacent to the chorion, the attachment is weak and easily disrupted during dissection. In the conceptus with normal attachment (KO #1, n = 1/4), less allantoic mesoderm is observed, and the resulting interface fails to expand as widely as in controls. | ||||
