Caption Placental phenotype in Bltp2em1(IMPC)J/Bltp2em1(IMPC)J (Bltp2 -/-) mice. At E9.5, Bltp2-/- placentas are properly developed and comparable to control littermates. At E10.5, Bltp2-/- placentas exhibit a diminished Tpbpa+ spongiotrophoblast (SpT) layer and moderate labyrinth zone (LZ) defects compared to controls. While the SpT layer abnormality is consistently observed in all Bltp2-/- placentas examined (n=4), the LZ defects are not fully penetrant, and their severity correlates with that of the embryonic phenotype. In one Bltp2-/- placenta, the LZ develops normally whereas in two of four Bltp2-/- placentas, the Tfeb+ labyrinth trophoblast layer is markedly reduced. Gcm1+ chorioallantoic branching is impaired in three of the 4 Bltp2-/- placentas.
