Placental defects in Alg3^em1(IMPC)J/Alg3^em1(IMPC)J (Alg3 -/-) embryos. At E9.5, Alg3 -/- placentas are properly developed and comparable to control littermates. At E10.5, Alg3 -/- placentas exhibit an expanded Tpbpa+ spongiotrophoblast layer and multiple labyrinth zone defects compared to controls. In knock-out placentas, the Tfeb+ labyrinth trophoblast layer is reduced in thickness but displays elevated Tfeb signal intensity, suggesting an increased density of Tfeb+ cells. Chorioallantoic branching is severely impaired as highlighted by the sparse Gcm1 expression. By E11.5, Alg3 -/- placentas display a more severe manifestation of the E10.5 defects.
