Transverse sections of WT (E-G) and double tail mutants (canp-dt; H-J) at the level of the first brachial arch (E and H), heart (F and I), and the level of the 14th somite pair (G and J), where the protrusion arose. In contrast to the neural tube in WT embryos, double tail mutants had a flat neural plate at the branchial arch level and a collapsed neural plate at more posterior positions. A notochord was visible below the neural tube in the wild sections (arrows in E-G), but no notochord was detected in mutant sections at the level of the branchial arch and heart. In WT sections, somites flanked the neural tube. By contrast, somites were fused across the midline in double tail embryos. A transverse section through the protrusion showed that it arose from the middle of the neural plate and was composed of mesenchymal cells (arrow in J). (Scale bars, 50 um in E-J.)
