Caption Loss of KATNAL1 function leads to reduced numbers of stable microtubules in Sertoli cells. (a) Western blot analysis and (b) quantification of stable (glu) and dynamic (tyr) alpha-tubulin on day 22 testes from wild-type (+/+) and homozygous Katnal11H/Katnal11H animals reveals a significant reduction in numbers of stable microtubules in testes from mutants (n=5-7). (c) Immunohistochemical localisation of glu-alpha-tubulin on corresponding day 22 testis sections localises stable microtubules to Sertoli cell cytoplasm (arrows) and developing spermatids and in wild-type animals. Conversely stable microtubules are below detection limits in much of the Sertoli cell cytoplasm in homozygous mutant testes. (WT1 = Wilms Tumour 1, SC-specific loading control).
