Light and electron micrographs show unique morphological changes in Crygb^Clapper/Crygb^Clapper and Crygb^Clapper/Crygb⁺ lenses. A: Light micrographs of the lens plastic sections show the fiber organization of 3 week old wild type (+/+), Crygb^Clapper/Crygb⁺ (I4F/+), and Crygb^Clapper/Crygb^Clapper (I4F/I4F) lens cross-sections around the lens equator, with the distances from lens capsule (50-150, 180-280, and 380-480 um, respectively). Some dark-stained droplets (indicated by arrowheads) appear only in the restricted inner region (180-280 um) of the homozygous lens section. Scale bars, 50 um for the left panels and 10um for all other panels. B: Electron micrographs show the interfiber spaces between the lens fibers of wild type, heterozygous and homozygous lenses with the distances from the equatorial capsule (60, 90, 120, 180, 330 um). Enlarged interfiber spaces can be seen in the region ~180-330 um from the lens capsule of heterozygous and homozygous lenses. C: The interfiber spaces were measured from the electron microscope micrographs in the areas 330-350 um away from the lens capsule. Statistical results show that the interfiber space in both heterozygous and homozygous lenses is ~3 times wider than that in wild type mice. (p<<-0.001). Scale bar, 200nm.
