Caption | In A-D, wild-type embryos are to the left and Sp(2H)/Sp(2H) mutant embryos are to the right. E-H show vibratome sections of the whole-mounts shown in C and D. Nonspecific trapping of probe can be seen in brain ventricles (A-D) and in the lumen of the neural tube in E. In A, expression is most intense in the ventral/posterior domain of the somites in the wild-type (arrows), whereas this domain is truncated ventrally in the mutant (between the small arrowheads). a, atrium of heart; da, dorsal aorta; nt, neural tube. In B, the most dorsal and ventral parts of the dermomyotome show expression (arrows), while expression is absent from the ventral part of the dermomyotome along the entire body axis in the mutant (arrowheads); expression is retained in thedorsal part of the somite at rostral levels of the body axis in mutant embryos. In C, transcripts are detected in a segmental pattern in the somites of wild-type embryos (arrows), whereas expression in the mutant is truncated ventrally (betweeen the small white arrowheads) and fused anteriorly-posteriorly (arrowhead). In D, discrete band of expression extend dorsoventrally in the wild-type (arrows), whereas these expression domains are truncated in the mutant (arrowheads). In E, expression is seen in the dermomyotome (between the small arrowheads) and in the sclerotome (large arrowheads). No expression is seen in the myotome (arrow). In a mutant sectioned at a similar level to E (F), the dermomyotomal expression domain (between small arrowheads) in truncated dorsoventrally, and the sclerotome (large arrowheads) expresses more intensely than in the wild type. G and H show sections at the level of the forelimb bud. Arrowheads show dermomyotome and arrows medial sclerotome. Scale bar: A, 1.1mm; B and C, 1.5mm; D, 2.2mm; E-H, 175 microns. | |||||||||||||||||||||||||||||||||||||||
