phenotype observed in males
N normal phenotype
respiratory system
• mice exhibit simplified alveoli (focal airspace enlargement) as early as P7 in the peripheral lung
• treatment with the VCP inhibitor, CB5083, improves alveolar morphology without causing overt toxicity
• however, no obvious fibrosis is seen in the lungs and autophagy and ER stress are not altered in alveolar 2 epithelial cells

phenotype observed in males
N normal phenotype
respiratory system
• mice exhibit alveolar simplification on P21
• alveolar 2 epithelial cells from tamoxifen-treated mice contain more mitochondria close to the cell surface than in controls

cellular
• maximal respiration capacity of alveolar 2 epithelial cells cell mitochondria is decreased and alveolar 2 epithelial cells show reduced glycolysis, glycolytic capacity, and glycolytic reserve, indicating mitochondrial dysfunction
• however, no differences in basal respiration are detected by oxygen consumption

N
• tamoxifen-treated adult mice show restoration of the decreased maximal respiration capacity of alveolar 2 epithelial cells cell mitochondria and improved glycolytic capacity that is seen in single mutant Sftpc mice
• tamoxifen-treated mice show enlarged lamellar body morphology in alveolar 2 epithelial cells

respiratory system
N
• tamoxifen-treated mice show restoration of the lung morphology seen in single mutant Sftpc mice, with rescue of the alveolar simplification in neonatal mice and in adult tamoxifen-treated mice
• tamoxifen-treated mice show enlarged lamellar body morphology in alveolar 2 epithelial cells
