cardiovascular system
• at E10.5, most homozygous embryos exhibit abnormal cardia tissue, some with enlarged hearts and some with pericardial edema
• however, heart development is normal at E9.5
• at E10.5, some homozygous embryos exhibit enlarged hearts
• at E10.5, some homozygous embryos exhibit pericardial edema
• at E9.5, some homozygous embryos exhibit abnormal blood deposition in the embryo or in the yolk sac
• abnormal blood deposition is often detected at E10.5
• at E10.5, some homozygous embryos exhibit abnormal blood deposition in the pericardial sac
• at E10.5, some homozygous embryos exhibit abnormal blood deposition in the midbrain

craniofacial
• at E10.5, nearly all homozygous embryos exhibit an abnormal head shape

embryo
• at E10.5, section analysis shows a reduction in cellular density in delayed homozygous embryos
• however, germ layer patterning is normal at E9.5-E10.5 and both heart and liver development are normal at E9.5
• at E9.5, some homozygous embryos exhibit incomplete turning
• at E10.5, most homozygous embryos exhibit abnormal branchial arch development
• a fraction of homozygous embryos are developmentally delayed at E9.5; nearly all are delayed at E10.5
• at E10.5, placentas exhibit multiple labyrinth zone defects
• at E10.5, the Tfeb+ labyrinth trophoblast layer is reduced in thickness but shows increased Tfeb signal intensity, suggesting an increased density of Tfeb+ cells
• at E10.5, yolk sacs dissected intact are vascularized but appear paler than in control littermates
• at E10.5, chorioallantoic branching is nearly absent, as Gcm1 expression is sparse and remains at the chorioallantoic interface
• however, placentas appear properly developed at E9.5

growth/size/body
• at E10.5, some homozygous embryos exhibit enlarged hearts
• at E10.5, nearly all homozygous embryos exhibit an abnormal head shape
• a fraction of homozygous embryos are developmentally delayed at E9.5; nearly all are delayed at E10.5
• at E10.5, heads sections are less cellularly dense relative to those in control littermates

homeostasis/metabolism
• at E10.5, some homozygous embryos exhibit abnormal blood deposition in the pericardial sac
• at E10.5, some homozygous embryos exhibit pericardial edema

mortality/aging
• homozygous embryos are present in Mendelian ratios at E8.5-E10.5; all homozygous embryos dissected at E11.5 are resorbing

nervous system
• at E10.5, some homozygous embryos exhibit abnormal blood deposition in the midbrain
