• at E5.5, TUNEL+ cells are specifically detected in the epiblast

embryo
• embryos fail to initiate gastrulation by E7.5, as shown by complete lack of T (brachyury) expression
• embryos do not progress morphologically past the E5.5 stage
• embryos do implant and form an egg cylinder but are developmentally delayed
• at E6.5, embryos are smaller but show normal pre-streak egg cylinder morphology
• at E7.5, embryos are much smaller than controls and show many morphological defects
• however, embryo size is normal at E5.5
• at E7.5, the embryonic portion is much smaller than that in control embryos
• at E5.5, the embryonic ectoderm appears slightly disorganized; however, epiblast cells properly express OCT4
• at E7.5, the epiblast largely remains as pseudostratified epithelium
• at E5.5, TUNEL+ cells are specifically detected in the epiblast
• at E5.5, the egg cylinder is slightly smaller but otherwise morphologically normal
• no morphologically or molecularly marked mesoderm is detected at E7.5
• no morphologically or molecularly marked primitive streak is detected at E7.5

• no exocoelomic cavity is detected at E7.5
• at E7.5, embryos show loosening/buckling of the visceral endoderm (VE); the VE appears to be losing contact with the epiblast and extraembryonic ectoderm resulting in a variable bubble-like cavity
• at E7.5, it appears as if the visceral endoderm has continued to proliferate

growth/size/body
• embryos do implant and form an egg cylinder but are developmentally delayed
• at E6.5, embryos are smaller but show normal pre-streak egg cylinder morphology
• at E7.5, embryos are much smaller than controls and show many morphological defects
• however, embryo size is normal at E5.5

mortality/aging
