• mice are viable and healthy and show no major changes in total cell number and composition of cell subsets (CD3+, CD4+, CD8+, B220+, Mac-1+, and Ly-6G+) in the thymus, spleen, mesenteric lymph node and bone marrow, indicating normal leukocyte development in lymphoid tissues
• mice exhibit normal alloreactive CD3+ T-cell proliferation in mixed lymphocyte culture assays, as determined by the % of CFSE-low cells within the CD3+ T cell population, and a normal delayed-type hypersensitivity (DTH) response to 2,4-dinitrofluorobenzene (DNFB), as determined by ear thickness at 24 and 48 hr after challenge
• when transgenic EL-4 lymphoma cells are injected intradermally into mice, 30%, 17%, and 82% of mice bearing Dd-, Kd-, and DdKd-EL-4 cells, respectively, reject the grafts on days 12 to 14 after transplantation, whereas wild-type EL-4 tumor cells are tolerated, indicating transgene number-dependent and MMR-independent rejection of Dd-, Kd-, and DdKd-EL-4 cells
• DdKd EL-4 lymphoma cells injected intraperitoneally into mice are rejected by TCRalpha beta+/CD8+ T cells, as seen in similarly treated wild-type mice
• surprisingly, peripheral blood mononuclear cells (PBMCs) and monocytes show loss of mRNA expression of both MMR1 and ATP9B (a precursor of MMR1), resulting in the tolerance to skin grafts from Dd or DdKd transgenic mice
• no binding of phycoerythrin-labeled H-2Dd pentamers to PBMCs is observed, unlike PBMCs from wild-type C57BL/6 mice which bind H-2Dd molecules with a very low Kd value
• mice fail to reject skin grafts from Dd, Kd, or DdKd transgenic C57BL/6 (H-2DbKb) mice
• in contrast, skin grafts from the B10D2 (H-2Id/minor Hb), BALB.B (H-2b/minor Hd), and C3H/He (H-2k/minor Hk) strains are rejected on days 12 to 14 after transplantation
