behavior/neurological
• the mean wakefulness bout duration is shortened
• however, the sleep/wakefulness pattern during the light period is not affected
• the transition frequency between wakefulness and sleep is increased in the first 3 hours of the dark period
• in the presence of DOX, mice show normal sleep/wakefulness patterns
• total amounts of slow wave sleep are increased during the first 6 hours of the dark period
• however, mean slow wave sleep bout durations are unaffected
• total amounts of REM sleep are increased during the first 6 hours of the dark period
• mice show severe fragmentation of wakefulness during the first half of the dark period, with a decrease in total time in wakefulness

mortality/aging
• 30% of mice survive beyond P120
• fewer mice die when over-expression begins at P60 instead of P40
• most mice die before 3 months of age frequently between P25 and P80
• in 37% of mice, crises of severe bradycardia and hypothermia progress to death
• however, 30% of mice survive beyond P120 and treatment with doxycycline prevents premature death

• mice exhibit sporadic autonomic crisis, involving bradycardia preceded 5 to 10 minutes by hypothermia, that persists for several hours and is followed by several days of recovery or, in the case of 37% of mice, progresses to death
• following treatment of brain slices with tryptophan, cells exhibit reduced firing compared to wild-type cells
• however, normal cell firing can be re-established by treatment with doxycycline
