Reduced body weight in Hap1tm2Xjl/Hap1tm2Xjl Tg(HCRT-cre)1Stak/0 mice
growth/size/body
• the greatest differences are seen at the ages of 2?4 weeks
behavior/neurological
• males and females consume 66 and 50%, respectively, of the food that control mice eat
• however, water consumption is normal
• show less activity than the control mice at night, with females showing a greater difference
• show less activity in response to fasting-induced metabolic stress
nervous system
• orexin neurons of the brain display disrupted or fragmented neurites in the dorsomedial hypothalamic and lateral hypothalamic nuclei

nervous system
N |
• despite loss of Gabbr1 expression, no decrease in the number of orexin positive neurons is detected and spontaneous excitatory postsynaptic currents in orexin neurons are similar to controls
• unlike in controls, treatment with bicuculline induces depolarization of orexin neurons
• membrane resistance of orexin neurons is significantly decreased
• glutamate and GABA-A agonists are less potent in inducing depolarizing currents in orexin neurons compared to controls
• increase in the frequency of fast spontaneous inhibitory postsynaptic currents in orexin neurons compared to control littermates
behavior/neurological
• severe fragmentation of the sleep wake cycle with decreased awake, non-REM, and REM sleep episode durations and reduced REM latency
• fragmentation occurs in both the light and dark phases
• the delta power is significantly increased in non-REM sleep and the theta power is decreased during REM sleep compared to controls
• however, the overall amounts of wakefulness, non-REM and REM sleep are similar to controls
