growth/size/body
• percentage of lean body mass is significantly lower compared to wild-type (at 13 weeks in males, and at 18 weeks in females)
• at 7 weeks of age, male and female transgenic mice show significantly reduced body weights compared to controls; by 22 weeks, reduction is 14% in males and 13% in females

adipose tissue
• mice have much less fat mass (40% of levels seen in controls)
• reduced by about half in females at 18 weeks

homeostasis/metabolism

• corticosterone, growth hormone, and insulin-like growth factor levels show no significant differences compared to wild-type (at 13 weeks)
• hypoleptinemia in mutants causes no changes in mRNA expression for neuropeptide (AGRP, POMC, NPY) in hypothalamus relative to wild-type
• responses to fasting, leptin treatment and deficiency are normal

• ablation of Pmch neurons is detected by postnatal week 4; by postnatal week 8, mice have lost 60% of Pmch expressing neurons in the hypothalamus and by postnatal week 15, 65% of these neurons are lost
