nervous system
• the fraction of pregnenolone sulfate (PS) sensitive neurons is drastically reduced in the dorsal root ganglia and trigeminal ganglia and PS stimulation fails to induce inward currents in patch clamp assays
• moderate but significant reduction in the heat sensitivity (43oC) of dorsal root ganglia and trigeminal ganglia neurons with a strong reduction in the population of cells responsive to both PS and heat
behavior/neurological
• during the first 30 minutes of a thermal preference assay mice spend more time at temperatures between 31 and 45oC compared to controls
• however, over the duration of the assay thermal preference is similar to controls
• injection of pregnenolone sulfate (PS) into the plantar skin of the hindpaw does not evoke any nocifensive behavior (paw licking and lifting)
• lack an aversive response to the presence of PS in drinking water
• response to hindpaw injection of capsaicin is normal
• injection of complete Freund's adjuvant fails alter hot plate withdrawal
• strongly increased tail flick latencies in a tail immersion assay and longer delays in a hot plate assay at 52 - 58oC
