Mutation details:
Arginine codon 177 (CGG) was changed to glutamine (CAG) (p.R177Q) and an FRT site flanked neomycin resistance gene cassette was inserted in to intron 2. The neo cassette was removed through subsequent Flp-mediated recombination. The mutation, in the cyclic nucleotide-binding domain of the encoded protein, is the equivalent of the same human mutation associated with early-onset thoracic aortic disease and in heterozygous mice leads to a similar phenotype.
