Arginine codon 498 (CGG) in exon 6 was changed to tryptophan (TGG) (p.R498W) using an sgRNA and an ssODN template with CRISPR/Cas9 technology. The mutation, in the highly conserved in the third extra-cellular LNS (laminin-neurexin-sex hormone binding globulin) domain of the encoded protein, replicates rs241832271C>T SNP found in the 129S1 strain and leads to enhanced empathy fear.
