Mutation details

Glutamine codon 71 (GAG) was changed to glycine (GGG) and glycine codon 72 (GGC) to serine (AGC) (p.71E_72GdelinsGS) using ZFNs and a donor plasmid template. The mutations, in the first zinc finger of the DNA-binding domain of the encoded protein, abolish the canonical binding of thyroid hormone to this receptor without affecting non-canonical interactions.
