Mutation details: CRISPR-targeting inserted 2 base pairs into the coding sequence (c.233_234insTT) in exon 4. This mutation results in a frameshift and premature termination (p.M78Ifs*2). It is based on the same human mutation found in azoospermic patients with meiotic arrest. RT-qPCR analysis of Redic1 expression at the cDNA level revealed no degradation of mRNA in testis tissues. Western blot of testicular lysates from 20 dpp homozygous mutant males showed absence of potentially in-frame reinitiation-translated proteins. Immunofluorescence staining showed no REDIC1 signals on chromosome axes of mutant spermatocytes.
