Mutation detailsThe following were inserted into intron 1, 130 bp upstream of exon 2: a loxP site, a copy of the last 130 bp of intron 1, wild-type cDNA coding for exons 2-5, human GH gene 3' UTR, 4x poly(A) signal sequences, an FRT site flanked neomycin resistance gene cassette, and a second loxP site. The neo cassette was removed through subsequent Flp-mediated recombination. Endogenous exon 2 was modified by changing glutamic acid codon 79 (GAA) to glutamate (CAA) (p.E79Q). The mutation is the equivalent of the same mutation found in human lung cancer. This allele expresses the wild-type protein and only after Cre-mediated deletion of the inserted cDNA will it express the modified protein. (J:353988)
