Mutation details: CRISPR-targeting of exon 6 deleted 5 bp. This deletion caused a frameshift in the Caprin2 ORF and generated a downstream premature stop codon in exon 7. The premature stop codon in the 7th exon of a total of 19 exons was shown to cause non-sense mediated decay of mRNA, resulting in much less expression of the truncated form. Western blot analysis showed that protein expression is significantly decreased in the eyes of homozygous mutant mice at P0.5. Immunostaining of P0.5 eye slices with an anti-CAPRIN2 antibody revealed a decrease in lens fiber cell staining.
