Mutation detailsThe transgene, inserted into chr11, contains the following elements: long-range Shh enhancer Rr29 (ZRS) upstream of mouse Hspa1 minimal promoter, mCherry reporter gene cDNA and SV40 poly(A) signal sequences (all in reverse orientation), a synthetic strong insulator, mouse Hspa1 minimal promoter, EGFP reporter gene cDNA and SV40 poly(A) signal sequences. Through short range activity of the enhancer, the allele expresses only mCherry in the embryonic limb buds; the insulator blocks the enhancer from controlling EGFP expression. (J:370789)
