Mutation details: ENU mutagenesis was used to induce a C to T mutation that is located within exon 6 at position 2:31,284,698 (ENSMUSG00000062661) and changes the proline at position 144 to a serine (P144S) in a region between the third and fourth EF-hand of the encoded protein. This point mutation introduces a third degree of freedom of rotation at residue 144 and potential for at least two additional hydrogen bonds that could interfere with proper protein folding and/or give rise to protein instability. Quantitative PCR showed no differences in whole brain mRNA levels whereas immunoblot analysis revealed a gene dose-dependent reduction in protein levels, consistent with the in-silico predicted destabilization of protein structure. Use of a different antibody confirmed the significant reduction of protein expression in hippocampal lysates from homozygous mutant mice.
