• Mutation details: Exon 2 was replaced with an mCherry fluorescent protein coding sequence followed by a floxed neomycin resistance gene cassette. The neomycin cassette was removed via Cre-mediated recombination in the germline. RT-PCR and quantitative PCR analysis confirmed the absence of Rnase2a transcripts in the lung and spleen of homozygous mutant mice after intranasal administration of IL-33. A Meso Scale Diagnostics (MSD) assay using antibodies raised against recombinant mouse (rm)Rnase2a confirmed the absence of protein in supernatants from macrophages stimulated in vitro with IL-4.
