Mutation details: This transgene comprises two copies of a modified version of an immunoglobulin kappa light chain (Igk) construct containing the rearranged mouse 2-phenyl-oxazolone (phOx) specific VkOx1-Jk5 variable region, the mouse intronic Igk enhancer, the rat Igk constant region (Igk-C) and 8 kb of mouse 3'-flanking sequence including the 3' enhancer. The modification consists of the introduction of two eleven-TG dinucleotide tracts, one at the border of complementarity determining region 1 (CDR1) and framework region 2 (FR2) of the variable region and the second at the beginning of Igk-C. These insertions cause a frameshift that creates a stop codon at the beginning of CDR2, so the gene as passed through the germline is non-functional. It undergoes mutation in the B-cell lineage, at least in Peyer's patches.
